标题: 流式通过Fluo-3 AM检测瞬时钙流遇到问题
利用Beckman 的Gallios机器检测淋巴细胞瞬时钙流遇到问题：
检测钙的染料为Fluo-3 AM，贮存浓度为5mM，工作浓度为5uM，激发光为488nm，发射光为525-530nm，步骤如下
提取人外周血单个核细胞，100ul体系，37°C避光孵育60min，PBS洗2遍，37°C避光孵育20min，立即上机。先跑1min作为荧光的基准值，立即加入CD3及CD28共刺激淋巴细胞，工作浓度均为10ug/ml，发现胞浆内的钙没有明显变化。图片见二楼

你也可以单用ionomycin刺激来验证前面染色和细胞活性是否没有问题。我感觉是CD3抗体刺激的问题哈。不过我是新鸟，期待高人回来

看图形钙离子没有被刺激出来，会不会实验步骤中还需要孵育？或者孵育用的缓冲液？
见下面1990年经典文章中的步骤：
PBL (10E7cells/ml) were incubated at 37°C for 20 min in HBSS containing fluo-3/AM in a final concentration of 4uM. Next, PBL were diluted 1/5 in HBSS containing 1% fetal calf serum and incubated for 40min at 37 ℃. The ceils were then washed three times and resuspended at a concentration of 1 x
10E6cells/mi in a Hepes-buffered saline, containing 137 mM NaCl, 5 mM KCl, 1 mM Na[sub]2[/sub]HP[sub]4[/sub], 5mM glucose, 1 mM CaCl[sub]2[/sub], 0.5 mM MgCl[sub]2[/sub], bovine serum albumin 1 g/l, Hepes 10 mM, pH 7.4.
Before each assay, the cells were incubated for 10 min in a 37°C water bath. The cells were subsequently stimulated with mAbs and lectins at the indicated concentrations and analysed on a Facscan flow cytometer

正好在做这个，我单用CD3也刺激不出钙流改变，但是后续加ionomycin阳性刺激还是有改变的。查了很多文献，说CD3的刺激需要cross linking 其抗体，我的是goat anti hamster IgG antibody。已经订了不过还没回来，不过还是不明白soluble CD3刺激需要cross linking其抗体，大侠解释一下吗？

Cross-linking each of these surface Ag with appropriate mAb and goat anti-mouse Ig (GaMIg) resulted in a unique pattern of increase in intracellular free calcium ([Ca2+]i) and different degrees of functional activation. 好像是这么回事，谢谢老师提供的文献！
