小白求教，看到文献里有报道在lin-CD117+ly6AE-筛选出的髓系祖细胞中，可利用CD16/32 和CD34区分出 CMP MEP 和GMP，不太明白CD34标出来的是什么样的细胞，在说明书上是这样解释的。CD34 Description: The RAM34 monoclonalantibody reacts with mouse CD34, also known as mucosialin. It has been reportedthat the RAM34 antibody can be used to detect CD34+Sca-1+c-Kit+ cells. CD34 isa highly glycosylated (approximately 90-120 kDa) member of the sialomucinfamily and is expressed by capillary endothelial cells,bone marrow stroma, and a small subpopulation of mouse bone marrow cells. RAM34 has been used to purify mouse hematopoietic stem cells (HSC) to nearhomogeneity. CD34 expressed on endothelial cells is a ligand for CD62L andplays a role in adhesion. Simultaneous staining of mouse bone marrow cells witha cocktail of antibodies to lineage markers (CD3, CD11b, Ly6G, TER-119 andCD45R/B220) reveals a subset of cells that stain with the RAM34 antibody andexpress undetectable to low levels of the indicated lineage markers.
